Líneas de Investigación

En medios biológicos las proteínas cumplen un rol fundamental en procesos metabólicos y estructurales (entre otros). El contenido de proteínas en células, tejidos y fluidos biológicos es significativamente superior al de otras macromoléculas, lo que, sumado a otros factores, hace que las reacciones de especies reactivas de oxígeno y nitrógeno con proteínas sean importantes en diferentes condiciones fisiológicas y patológicas. De hecho, la oxidación de proteínas se ha asociado directamente con la etiología de diferentes enfermedades neurodegenerativas y cardiovasculares.

Por otra parte, la oxidación de proteínas también es relevante en ámbitos alimenticios. Alimentos ricos en su contenido proteico son fundamentales para la nutrición humana y animal; la oxidación de proteínas en tales sistemas, por ejemplo, durante su procesamiento, es un fenómeno que puede tener consecuencias nutricionales e incluso toxicológicas. La oxidación de proteínas es un proceso complejo que involucra diferentes rutas de reacción lo que conlleva a la formación de múltiples intermediarios reactivos y productos. Tales procesos conjuntamente con alterar de manera importante la funcionalidad de las proteínas pueden llevar a la formación de productos oxidados no deseados.

Nuestro grupo de investigación (Free Radicals and Protein Oxidation) tiene como objetivo principal estudiar, desde una perspectiva amplia, la química de la oxidación de proteínas mediada por especies reactivas. Pretendemos entender los mecanismos de reacción de las proteínas con particular énfasis en los procesos mediados por triptófano y tirosina, y que llevan a cambios en la masa molecular de proteínas de relevancia nutricional y biológica. Para abordar tal desafío nuestro grupo ha mantenido colaboraciones con investigadores pioneros en el área, expertos en estudios computacionales, bioquímica, fotoquímica, físico química y biología.

Desde sus inicios, nuestra investigación ha sido financiada por proyectos Fondecyt lo que nos ha permitido implementar nuestro laboratorio con instrumental de alto valor como son equipos cromatográficos (HPLC y cromatografía de exclusión por tamaño), espectrofotométricos (fluorescencia y UV-visible; Lector microplacas) y electroforético (SDS-PAGE, western blotting con Trans-Blot Turbo). Adicionalmente, a través de un proyecto Fondequip, contamos con acceso a un equipo UPLC con detección de masas.

                                             


Pretendemos, a través de la comprensión de los fenómenos básicos de la oxidación de proteínas, aportar al conocimiento científico para la búsqueda de soluciones a diferentes patologías y problemas alimenticios asociados con procesos oxidativos. Nuestra experiencia previa en la química de radicales libres y de compuestos polifenólicos nos permite abordar los objetivos actuales desde una mirada científica particular. En un futuro esperamos avanzar en el entendimiento de reacciones oxidativas sobre proteínas que cumplen funciones importantes en rutas metabólicas relacionadas con el desarrollo de ciertas patologías. Adicionalmente esperamos comprender y proponer soluciones a desafíos actuales en torno a la industria alimentaria (Polos Estratégicos).
